I tried mplayer, which I'd much prefer to use: no luck. But VLC will work, and is recommended somewhere here. Google it to find the site with free download. Then you need to hit "browser" at the bottom of your "Directories" in Preferences (hmmm ... that location is intuitive, isn't it) and insert the VLC application. Then it should work by option-clicking on the progress bar of the incomplete file that you wish to preview.

Why, I wonder, isn't this information easy to find, since ALL Mac users want to preview files. It took me months to realise that you have to do this. ?(

Preview works fine with any player: vlc, xine, mplayer.

Do you have error messages in the terminal ?

No, I've just tried making Mplayer the preferred application (in Directories)—then it opens but the file won't play. So ... back to cruddy ol' VLC, which is seriously bad for the Mac—no speed control, bad at dropping in halfway through the file, etc. PS No error messages.

If you had actually read the README.Mac.txt file that comes with every Mac binary of aMule, you would have seen a whole section called "Setting up aMule's video preview feature" -- can't get any more obvious than that. ;)

spacecadet:
Did you use the instructions from the above readme file to set up mplayer with aMule? I'm pretty sure they worked when I wrote them (I personally prefer vlc for previews so I've tested mplayer only once or so). I can't do any testing right now because I'm not in front of my mule machine, but I'll try to remember to do some testing later.
